چکیده

The metabolism of chromatin proteins of rat liver has been examined during the prereplicative period following partial hepatectomy. Within 8 hours after partial hepatectomy, isolated chromatin exhibits a 7% decrease in its histone to DNA mass ratio compared to sham-operated and zero time controls. The time course and amount of this decline correlate well with the previously published pattern of chromatin template activity increase. During the decrease in histone, the relative amounts of the five major species remain constant. However, there are differential losses and resyntheses of certain histone species. Histone I shows the most pronounced turnover, an estimated 12.5 %. During regeneration, the non-histone chromatin protein to DNA mass ratio does not change significantly, nor do the electrophoretic patterns of major non-histone polypeptides. However, during the initial period of chromatin template activity increase (from 2% to 4 hours after the operation), the non-histones as a group exhibit an increase in their rate of labeling greater than that of other subcellular fractions. Moreover, the rates of labeling of many non-histone species become differentially stimulated, suggesting that their rates of loss from chromatin have also selectively increased.

Early and late changes in nonhistone chromatin proteins accompanying rat liver regeneration.

Chromatin was isolated from 0.025 M citric acid nuclei of regenerating rat liver at 1,5,18,24, and 48 hr posthepatectomy. The total protein to DNA ratios did not change significantly during this time period. However, 2-dimensional polyacrylamide gel electrophoresis of nonhistone proteins of "Chromatin Fraction II" revealed changes in the amounts of some protein spots. As early as 1 hr after hep...

Nuclear Architecture and Epigenetics of Lineage Choice

Differentiation is an epigenetic process which is installed by changes of transcriptional programs over successive cellular divisions. A number of studies have reported the effects of biochemical modifications of chromatin (DNA and chromatin proteins) on the regulation of transcription. Although, these studies are able to explain how transcription of a given gene is regulated (toward activation...

Mitochondrial ultrastructure & release of proteins during liver regeneration.

BACKGROUND & OBJECTIVES It has been reported that some proteins are released from mitochondria during liver regeneration after partial hepatectomy (PH), but the relationship between proteins release and mitochondrial permeability transition (MPT) remains unclear. We undertook this study to demonstrate the changes of mitochondrial ultrastructure and proteins release during liver regeneration and...

Id2 leaves the chromatin of the E2F4-p130-controlled c-myc promoter during hepatocyte priming for liver regeneration.

The Id (inhibitor of DNA binding or inhibitor of differentiation) helix-loop-helix proteins are involved in the regulation of cell growth, differentiation and cancer. The fact that the molecular mechanisms of liver regeneration are not completely understood prompted us to study the fate of Id2 in proliferating liver. Id2 increases in liver regeneration after partial hepatectomy, following the e...

Antigenic changes in nonhistone proteins during azo dye hepatocarcinogenesis.

We have investigated the appearance of specific nonhistone proteins during azo dye-induced hepatocarcinogenesis in the rat. Groups of animals fed azo dye-containing diet were sacrificed at approximately 3-week intervals, portions of their livers were examined histologically, and the remaining material was fractionated into chromatin and cytoplasmic fractions. Livers of the azo dye-fed animals e...
